RESUMO

Objective:To explore the effect of physical development on thyroid volume of children aged 8 - 10 years in Sichuan Province, and explore the thyroid volume correction method suitable for school-age children, so as to accurately prevent and control iodine deficiency disorders.Methods:From June to July 2020, Shuangliu District of Chengdu City, Pengshan District of Meishan City, Miyi County of Panzhihua City and Qingchuan County of Guangyuan City were selected as the survey counties (districts). One township (town and street) was selected from each county (district) according to the five directions of East, West, South, North and Middle, one primary school was selected from each township (town and street), and 40 children aged 8 - 10 years (gender and age balanced) were selected as the survey subjects from each primary school, height and weight were measured, the body mass index (BMI) and body surface area (BSA) were calculated. Thyroid volume was measured by B-ultrasound, and the different thyroid volume indexes [height volume index 1 (HVI1), height volume index 2 (HVI2), body mass volume index (BMIV), weight and height volume index (WHVI), body surface volume index (BSAV)] were calculated, respectively. Urine samples of all children were collected, the iodine concentration in urine was measured, and the correlation between different measurement indexes and children's growth and development indexes was analyzed.Results:A total of 805 children aged 8 - 10 years were investigated, including 403 boys and 402 girls. There were 312, 288 and 205 children in the 8-, 9- and 10-year-old groups, respectively. A total of 805 urine samples were collected, and the median urinary iodine was 251.4 μg/L. There was no statistically significant difference in thyroid volume between boys and girls ( Z = - 0.44, P = 0.661), but was statistically significant difference between ages ( H = 64.95, P < 0.001). In all age groups, thyroid volume was positively correlated with height and weight (8-year-old group: r = 0.29, 0.42, P < 0.001; 9-year-old group: r = 0.29, 0.41, P < 0.001; 10-year-old group: r = 0.34, 0.47, P < 0.001). In all age groups, after HVI1 correction, thyroid volume was positively correlated with height and weight (8-year-old group: r = 0.13, 0.32, P < 0.05; 9-year-old group: r = 0.12, 0.30, P < 0.05; 10-year-old group: r = 0.18, 0.37, P < 0.05). In all age groups, there was a positive correlation between thyroid volume and weight after HVI2 correction (8-year-old group: r = 0.20, P < 0.001; 9-year-old group: r = 0.17, P = 0.004; 10-year-old group: r = 0.26, P < 0.001). In the 8- and 10-year-old groups, there was a positive correlation between thyroid volume and height after BMIV correction ( r = 0.20, P < 0.001; r = 0.21, P = 0.003). In all age groups, there was a negative correlation between thyroid volume and height and weight after WHVI correction (8-year-old group: r = - 0.35, - 0.37, P < 0.001; 9-year-old group: r = - 0.38, - 0.39, P < 0.001; 10-year-old group: r = - 0.31, - 0.38, P < 0.001). In the 8-year-old group, there was a positive correlation between thyroid volume and weight after BSAV correction ( r = 0.11, P = 0.045). Conclusions:Thyroid volume is closely related to height and weight. It may be inappropriate to judge goiter in children only based on age. After the preliminary comparison of five correction methods, it is found that BSAV is better.

RESUMO

N 6 -methyladenosine (m6A) is a chemical modification that exists in a variety of RNAs and is most commonly observed in mRNA. The liver is a vital metabolic and digestive organ in human body, and m6A methylation plays an important role in the physiological and pathological processes of the liver. This article reviews the biological role and potential application value of m6A methylation in liver physiology and liver diseases such as viral hepatitis, nonalcoholic fatty liver disease, liver fibrosis, and hepatocellular carcinoma, and it is pointed out that m6A methylation can regulate related factors and is involved in the development and progression of liver diseases, which provides new ideas and targets for clinical diagnosis and treatment.

RESUMO

AIM: To observe the protective effects of Xiaochaihu Decoction (XCHD) on carbon tetrachloride (CCl4)-induced hepatic fibrosis (HF) in mice. METHODS: HF was induced in male mice by 20% CCl4 twice a week for 12 weeks. XCHD (3.90, 7.80, 15.60 g/kg) and Silybin (0.1 g/kg) were administrated via gavage once a day starting from the CCl4 treatment for subsequent 12 weeks. Then, the levels ALT and AST in serum were assayed, liver samples were taken to examine the degree of HF by HE, Masson, Sirius Red staining and Electron microscope. Moreover, the protein and mRNA expression levels of α-smooth muscle actin (α-SMA) and Collagen Ι were determined by RT-qPCR, immunofluorescence and Western blot. RESULTS: The data demonstrated that XCHD (7.80, 15.60 g/kg) effectively reduced histopathological changes, such as steatosis, collagen deposition; meanwhile the histopathological analysis suggested that XCHD obviously alleviated the degree of HF. Furthermore, XCHD significantly reduced the levels of ALT, AST (P<0.05), and attenuated the expressions of α-SMA, Collagen Ι both of protein and mRNA levels (P<0.05). CONCLUSION: XCHD has protective effect on chemical HF in mice.

RESUMO

Hepatic fibrosis is a reversible pathological reaction of the liver during the self-repair of various chronic liver injuries and can progress to liver cirrhosis and liver cancer. Pyroptosis is a new type of programmed cell death different from apoptosis and necrosis and is closely associated with the pathogenesis of hepatic fibrosis. This article reviews the latest research advances in the definition of pyroptosis, related activation pathways, and its role in hepatic stellate cells, hepatic parenchymal cells, Kupffer cells, and eosinophils, and it is pointed out that pyroptosis can induce liver inflammation, excessively activate hepatic stellate cells, and promote the development and progression of hepatic fibrosis, in order to provide new ideas and potential targets for clinical diagnosis and treatment.

Objective To study the predictive validity of fidgety general movement assessment in pre-term twins and multiplets for motor development outcomes.Methods A total of 53 pre-term twins or multiplets delivered between July 2011 and February 2016 participated in this study.They were assessed using a general movements (GM) assessment and participated in the follow-up program until one year old.The motor development outcomes of the infants at one year old were determined according to clinical diagnoses and the Peabody developmental motor scale number two (PDMS-2) evaluation.The predictive validity of fidgety general movement assessment for motor development outcomes was calculated against the standard motor development of infants at one year old.Results There were 53 twins or multiplets who accepted the GM assessment of fidgety movement period.Of these,43 were assessed as normal (NF) and ten (19%) as lacking a normal level of fidgety movement (F-).All 53 cases were followed-up for the motor development outcome.Forty-three cases (81.1%) were assessed as normal at one year old,while ten (18.9%) were assessed as abnormal.All ten had cerebral palsy,and no motor development retardation was found.The predictive value of F-for cerebral palsy was 90.0% in terms of sensitivity,97.7% in terms of specificity,90.0% in positive predictive value,and 97.7% in terms of negative predictive value.Conclusions Among pre-term twins or multiplets,the fidgety general movement assessment can be a useful early indicator of motor development difficulties.

Objective To observe the repeat administration toxicity of Guanxinning Injection in rats.Methods Totally 120 rats (males and females) were randomly divided into vehicle control group and three Guanxinning Injection groups with different dosages (3.40,1.70,and 0.85 g/kg).Rats were administered with Guanxinning Injection by consecutive intravenous injection for 13 weeks.Besides the general conditions were observed,the related indexes were detected,such as body weight,the routine control of blood,hepatic function,renal function,the metabolism condition of lipids,the glycometabolism indexes,and histopathology analysis were determined at 13 weeks of treatments and 4 weeks after the withdrawal,respectively.Results Rats in the Guanxinning Injection group at 3.40,1.70 g/kg apperance shortness of breath,unsteady gait,lying motionless,and other symptoms.There was no obvious abnormal reaction in the 0.85 g/kg dose group.There were two male rats in the Guanxinning Injection group at 3.40 g/kg died at about week 4 of treatments,and there was no death in the 0.85 and 1.70 g/kg dose groups.Compared with vehicle control group,the related indexes of blood,blood biochemistry,organ relative quality,and histopathological showed no obvious abnormalities in Guanxinning Injection 0.85 and 1.70 g/kg group.The levels of urea nitrogen (BUN) and the relative weight of kidney in Guanxinning Injection 3.40 g/kg group were significantly higher than those in vehicle control group at 13 weeks.The change of these parameters regained to normal at 4 weeks after withdrawal and the rest of the detection index showed no obvious abnormality in 3.40 g/kg dase group.Conclusion Intravenous administration of Guanxinning Injection for 13 weeks at high dose could induce reversible damage to kidney.

<p><b>OBJECTIVE</b>To investigate the effect of amphotericinB (AmB) on migration and invasion of esophageal carcinoma Eca109 cells exposed to hypoxia and explore the molecular mechanisms.</p><p><b>METHODS</b>Routinely cultured esophageal carcinoma Eca109 cells were treated with 0, 1.25, 2.5, or 5 µg/ml AmB in hypoxic condition (3% O2, 5% CO2, and 92% N2) for 24 h. The cell migration and invasion were assessed by cell scratch test and Transwell chamber assay, respectively. Real-time quantitative PCR and Western blotting were used to detect the mRNA and protein expressions of hypoxia-inducible factor-1α (HIF-1α), matrix metalloproteinase-2 (MMP-2), and E-cadherin in the cells, respectively.</p><p><b>RESULTS</b>Compared with the control cells, the cells treated with different doses of AmB showed attenuated ability of migration and invasion (P<0.05). AmB treatment resulted in significantly lowered mRNA and protein expressions of MMP-2 (P<0.05) and increased expressions of E-cadherin (P<0.05); the protein expression of HIF-1α decreased significantly in cells after AmB treatment (P<0.05) but its mRNA levels showed no significant changes (P>0.05).</p><p><b>CONCLUSION</b>AmB can suppress the migration and invasion of esophageal carcinoma Eca109 cells in hypoxic microenvironment possibly by regulating the expressions of HIF-1α, MMP-2 and E-cadherin.</p>

RESUMO

Objective To study the compressive education on nursing students in operation room.Methods 84 nursing students in practice were randomly divided into the observation group and the control group with 42 students in each group.The two groups were in a month to complete the teaching syllabus internship period.The control group received conventional teaching,on the basis of conventional teaching,the observation group was given compressive education.After the end of internship,nursing skill score,comprehensive quality,operation doctors'satisfaction degree were compared between two groups.Results After the compressive education,the nursing skill score,comprehensive quality,operation doctors'satisfaction degree in the observation group were significantly higher than those of the control group.Conclusions According to the source of pressure in the operation room,compressive education should be given to the nursing students and make them complete the practice in a relaxed and delight state.This can also reflect the humanistic care in teaching,which is worthy of being popularized in teaching in operation room.

Objective To investigate the characteristic and its clinical value of tumor necrosis factor (TNF)-α and its receptor, interleukin (IL)-1β and its receptor in serum and bronchoalveolar lavage fluid(BALF) in patients with pulmonary tuberculosis and to determine the role of them in the immunopathogenesis of tuberculosis. Methods The concentrations of TNF-α,soluble TNF receptor (sTNF-R) Ⅰ, IL-1β and IL-1 receptor were measured using sandwish ABC-enzyme-linked immunosorbent assay (ELISA) method in serum and BALF of 46 patients with active tuberculosis and 21 patients with inactive tuberculosis, and in the serum of 20 cases of healthy control. Meanwhile the above-mentioned cytokine levels in serum and BALF of 19 patients with active tuberculosis were followed up. Differences between groups were assessed for significance by t test. Results The TNF-α,sTNF-R Ⅰ, IL-1β and IL-1 receptor levels and TNF-α/sTNF-R Ⅰ ratios in BALF of active tuberculosis group were (286.2±96.3) pg/L,(2 431.5±1 124.6) pg/L,(58.6±3.2) pg/L,(162.4±17.1) pg/L and 0.06±0.01, respectively, which were all significantly higher than those with inactive tuberculosis group (t=3.36,3.25,2.95,2.27 and 3.12 respectively; P<0.05). The TNF-α,sTNF-R Ⅰ,IL-1β and IL-1 receptor levels and TNF-α/sTNF-R Ⅰ ratios in BALF of cavernous tuberculosis group were (381.4±106.4) pg/L,(2 824.7±1 318.5) pg/L,(66.4±4.6) pg/L,(176.4±18.7) pg/L and 0.07±0.01, respectively,which were all significantly higher than those of non-cavernous tuberculosis group (t= 3.46,2.37, 3.19, 2.99 and 3.22, respectively; P<0.05). After 2-month' antituberculosis treatments, among 19 cases, the TNF-α,sTNF-R Ⅰ,IL-1β and IL-1 receptor levels and TNF-α/sTNF-R Ⅰ ratios in BALF of 16 cases were significantly lower than those at the beginning of treatments (t= 3.26,3.17, 3.28, 2.92 and 3.12 respectively; P<0.01). Meanwhile, their clinical symptoms improved, sputum smear negative, lesions on chest X-ray resolved and the cavity shrinked or closed. Conclusions TNF-α, sTNF-R Ⅰ, IL-1β and IL-1 receptor are likely to be involved in the immunopathogenesis of tuberculosis. Detection of TNF-α, sTNF-R Ⅰ, IL-1β and IL-1 receptor levels in the serum and BALF is helpful to understand the activity of disease, determine the clinical pattern of disease,assess the prognosis of disease and monitor the therapeutic effect in patients with pulmonary tuberculosis.

Objective To discuss the growth suppressing, apoptosing effect of new type tumor-supressor gene-p33ING1 in stomach cancer cell strain, and to explore new strategies and methods in tumor therapy. Methods The PCDNA3/p33ING1 nuclear expressing microsome was constructed, p33ING1 and wt-p53 were implanted to human stomach cancer cell both and to evaluate the effect of p33ING1 and p53 toward stomach cancer cell and synergism between them. Results The PCDNA3/p33ING1 nuclear expressing microsome was successfully constructed. The human stomach cancer cell strain SSCG-7901 under implantation of p33ING1 and wt-p53 showed a significant decrease in cell growth, the coupling time was delayed, DNA synthetic phase was shortened and G0/G1 phase prolonged. The cell collapse increased. Conclusions Despite of the tumor-inhibiting effect and biochemical activation of p33ING1, it also plays a role with p53 gene in controling growth of stomach cancer cell, inducing cell collapse and hampering cell proliferation cycle. P33ING1 and p53 are synergistic to each other.

RESUMO

AIM: To develop a method to detemine pinostrobin in Weitengning Talets (Lindera reflexae Hemsl.) by HPLC. METHODS:A C_(18) column was used,methyl alcohol-water(80∶20) was used as a mobile phase and the wavelength of UV detector was set at 290 nm. RESULTS:The linearity of this method was good with the average recovery of 97.9%,RSD was 0.74%(n=5). CONCLUSION:The methed is simple,reliable,sensitivity,and with good reproducibility.It can be used in quality control of Weitengning Tablets.
